What is "t-" and "t+" mean...

Dear VPI,

I've heard about and seen pictures of a lot of snakes, mostly albino retics, and some are marked t+ and some aren't.

What exactly does it mean when it said they are “t+”?Thanks, kevin  

 Dear kevin,

"t+" means "tyrosinase-positive" and "t-" means "tyrosinase-negative." 

Tyrosinase is an enzyme that acts on tyrosine, an amino acid that is the basic building block of melanin and a suite of other dark pigments that are called melanin-related pigments.

A tyrosinase-negative albino is an albino that does not have functioning tyrosinase or has a defective tyrosinase that does not work correctly, and so no black melanin can be produced. Also no melanin-related pigments are produces.  A tyrosinase-positive albino has functional tyrosinase and it is able to complete the first and second steps of the multi-step process of black melanin synthesis. Some other enzyme along the synthesis is missing. Tyrosinase-positive albinos do not make black melanin, but they do make other melanin-related pigments. Melanin-related pigments are mostly dark colors and they include a purple-gray, dark gray, gray brown, a dark yellowish brown, and several shades of brownish-red, dark red or medium red.

 Examples of t- albinos would be albino ball pythons and albino Burmese. T+ albinos would include Bob Clark's albino retic, caramel-albino ball pythons, and VPI's lineage of red-albino blood pythons.   DGB

I was watching a ralph davis video on youtube and he had two Caramel albino's with kinks in them, And ive read up You get quite alot of kinks in caramel albinos with visual to visual breeding? Is this true?

To minimise kinking, breed unrelated from different lines. The majority of kinked Caramels are resulting from breeding closely related parents.

its "one" of the ball python hobbys dirty secrets, ralph made a big thing about speaking about the caramel kinks, but he's obviously known for a while and never said anything when they where fetching big bucks, no one did, it would affect thier sales, it was only when general hobbyists got them that it was spoken openly about, obvious others are the spider wobble and the super's duck bill, strange how people call these problems "cute" in ball pythons, if it was a corn, it would be put it down, thats awful etc etc, but balls and money go hand in hand so for now at least these arent problems, thier "cute" side effects.

Breeding hets can also decrease the kinking, kinked ones well some get kept as show things but most end up in the freezer to be blunt...

Caramel Albino

The Caramel Albino, also known as Xanthic or T+ Albino, is one of the most unique looking Ball Python morphs out there. The coloration is caused by a small amount of Tyrosine (Melanin) more than a regular albino resulting in Purples and Yellows. You don't see too many of these around. My original het Caramel female just ovulated (the female that produced the regular albino a couple years ago). She was bred by my Pastel het Caramel.......hopefully she produces something other than normals!

The first regular albino (T-) produced in Canada was produced from breeding 100% het Caramel (T+) to a 100% het Caramel (T+). That is why this year I bred my Pastel het Caramel to my original het Caramel (I have a couple het caramels). That way if I don't produce caramels I know I got screwed.

My female is fine. Although, almost every breeder in the USA who has produced Caramels in numbers has produced kinked offspring. It's in the gene and it doesn't seem to be related to inbreeding........although I'm sure it's not helping. Breeders in the USA who are working with totally unrelated stock (wild caughts) are still producing kinked offspring. Most of the kinking seems to be in the mid section (severe spinal kinks) although some show up in the tail and aren't as bad. 

Problematic Caramels and spiders are not something people like to talk about as it hurts their bottom lines.

money for mutants (genetic defects). I, for one, am all for the morphs. Even though there are some wild caught albino ball pythons floating around, there are only very few of them. If you have an albino ball python, odds are that you purchased a captive-bred animal. For the most part, I think it promotes captive bred animals (let's not get into the huge demand for female ball pythons though - another subject).

What gets really complicated, is when is the defect more than we bargained for? Typically, pigmentation or pattern mutation are not evaluated as problematic for the animal in any way. And for the most part, we don't have any issues with that. One could still argue that for an animal that is not nocturnal, albinism could be not that great... but let's not go there since we don't have that problem with Ball Pythons heh...

However, when talking about an interesting pattern and/or color mutation that is accompanied by some unwelcomed defects, the question is "when is it ok or not ok to breed them"? There is a gray zone here. Discussions in this zone is where people get all fired up. The reason is that not everybody agrees what is too much and what isn't.

In my opinion, and that's all it is, there should be a way to determine if the defect is serious enough or not to terminate a particular project. I think that any physiological or mental defect that is problematic for the animal's "well being" should be enough to terminate a project. That would include skin problems (rashes, thin skin that rips open easily, etc), problems eating, problems defecating, problems with or does not breed, if it was possible to know it had pain in some way, had a shortened life span for some reason, mobility was restricted, some particular sense does not work (such as being blind), and so on...

An animal's well being should be defined I suppose. What do ball pythons do in the wild? When is a ball python comfortable and "happy" (if that can be said of a ball python)? All they do is move around, eat, breed and defecate. If a snake is capable of doing all that without any pain or physical discomfort, I see no reason why not to breed it. One might say, a snake can do all of that and be blind! Well the snake can, but I'm convinced the stress level of that snake would go up considerably. A blind snake would not be a snake to breed in my opinion.

Corey's decision to not breed kinked caramels is the only respectable approach, because in this particular case, we know it is somehow related to the caramel genetics and it is not a incubation-related problem. Perhaps at some point we may be able to "breed it out", who knows? There are a lot of Caramels that are not kinked. I would think this means that there may be a possibility of breeding it out. Let's all hope that will happen. Breeding snakes for the freezer is not what we're all into this for.

For other particular defects, such as the spinning spiders, not all agree on what should be done about it. Everybody is entitled to their own opinions on it. This certainly falls into the "gray zone" I was talking about. It looks like more kinked caramels are going to be sent to the freezer than spiders because with the caramel, as Corey stated, it's obvious at birth. The spinning spiders aren't as obvious to see at birth. In my opinion however, the "spinning spider syndrome" is not anything that affects the snakes "quality of life" in any way. A LOT of breeders have stated that their spiders were eating & breeding machines.

So, that's my take on it... I talk too much, sorry for the long post... hopefully it will make some sense to somebody :D

Dominant suppression = A form of epistasis in which a dominant allele at one locus suppresses the effect of a dominant allele at another locus (Klug, 2003).
Dopa = 3, 4-dihydroxy-L-phenylalanine, an intermediate chemical in the synthesis of melanin (Bechtel, 1995).
Dorsal = Pertaining to the back; an anatomical direction meaning toward the back (Barker, 2006).  The top, back, or uppermost surface of an organism (Bechtel, 1995).
Dorsolateral = Pertaining to the area between the dorsal and lateral surfaces (Barker, 2006).
Dorsum = The upper surface or back of a python (Barker, 2006).
Double Heterozygous = being heterozygous for two different traits.
Ectopic expression = Gene expression that occurs out side the cell or tissue where the gene is normally expressed (Hartwell, 2004).
Ectothermic (Poikilothermic or cold-blooded) = Possessing no internal means for controlling body temperature (Bechtel, 1995).
Embryo = Organism in early stages of development, before it is physiologically independent (Bechtel, 1995).
Endothermic (Homeothermic or warm-blooded) = Having ability to maintain constant body temperature in different ambient temperatures (Bechtel, 1995).
Epistasis = The situation occurring when one allele of one gene obliterates the phenotypic expression of all other allelic alternatives of another gene (Suzuki, et al., 1986).  A gene interaction in which the affects of an allele at one gene hide the effect of alleles at another gene (Hartwell, 2004).  Nonreciprocal interaction between genes such that one gene interferes with or prevents the expression of another gene (Klug, 2003).
Erythrism = Having red skin and scales caused by a lack of black pigments (eumelanin), which allows the red pigment (pheomelanin) to dominate the color of the appearance (Allaby, 1991).  Abnormal or excessive amount of red coloring (Holmes, 1979).  The occurrence of unusual amounts of redness in an individual or population as compared to the normal pattern of the species (Peters, 1964).

Erythrocystic =  See erythrism.
Erythrophores = Reddish-purple pigment-bearing cells (Holmes, 1979).  Cells containing carotenes or yellow pigment(Bentley, 1982).  Xanthophores that appear red (Bechtel, 1985).
Erythrophore = Red chromatophore (Bechtel, 1985).
Eumelanin = A form of melanin that is black or dark brown (Mattison, 1986).  Black or brown melanin (Bechtel, 1985).  See phaeomelanin.
Triple Heterozygous = Heterozygous at three gene loci.

P generation = Two unlike individuals that begin a genetics experiment, or breeding program.
F1 generation = First filial generation; the offspring of the P generation.  An F1 is a single member of the F1 generation.
F2 generation = Second filial generation; the offspring of two F1s.
F3 generation = Third filial generation; the offspring of two F2s.
Gene = Unit of heredity that determines the characteristics of the offspring.
Genetics = The study of heredity.
Genotype = An organism’s genetic composition. Heredity - the transmission of genetic characters from parents to offspring.
Het = An abbreviation for heterozygous.
Heterozygous = Possessing two different genes for a given trait. An animal with one mutated, recessive gene still appears normal; its mutated gene can be inherited by future offspring. A co-dominant animal is heterozygous for the dominant form of its mutated gene, yet is different in appearance than both the wild-type and homozygous forms.
Homozygous = A state in which both genes for a specific trait are the same. When a recessive gene is it its homozygous form, it makes the animal look different from the wild-type. When a dominant gene is in its homozygous state, it causes the animal to look different from both the wild-type and the heterozygous (co-dominant) forms.
Hypomelanistic = An animal having less black and/or brown color than a wild-type.
Leucistic = A pure white animal with dark eyes.
Locus = A gene's position on a chromosome (plural: loci)
Melanin = Black or brown skin pigments.
Melanistic = Abnormally dark, especially due to an increase of melanin.
Mutation = An abnormal gene that under certain circumstances can cause an animal to be born with an appearance other than wild-type.
Normal = An animal with no mutated genes - "wild type" in appearance. (See wild type)
Phenotype = An animal’s external appearance, as caused by its genotype.
Possible Het = An animal from a known breeding that has either a 50% or 66% possibility of being "heterozygous" for a mutant gene.
A 66% possible het comes from breeding 2 heterozygous animals together: 50% of the offspring are heterozygous, 25% will be homozygous, and 25% will be wild-type. Of the normal appearing animals, 66% (or roughly 2 out of 3) will actually be heterozygous for the mutated gene.
A 50% possible het comes from breeding a heterozygous animal to a wild-type animal. All of the resulting offspring will be wild-type in appearance, but 50% of them will actually be heterozygous for the mutated gene and must be bred out to determine which animals are really hets.
Punnett Square = A learning tool for determining the possible outcomes of a given cross between individuals.  It was developed by R.C. Punnett, an early British geneticist.
Recessive = A gene that affects an animal's appearance if it's present in the homozygous state. A heterozygous animal carrying a mutated, recessive gene looks normal. Some examples of this would be Albino, Ghost/Hypo, Caramel Albino, Axanthic, Piebald, Clown, Desert Ghost, Orange Ghost, Hypo, Genetic Stripe andLavender Albino Ball Pythons.
Super = A Commonly used Herpetocultural term for the Dominant form of a Co-dominant mutation, I.E. Super Pastel.
Tyrosinase = An enzyme required for synthesizing melanin.
Tyrosinase-negative = An albino whose cells lack tyrosinase, producing a white and yellow/orange animal with pink eyes. A separate albino mutation from tyrosinase-positive. Also called T- .
Tyrosinase-positive = An albino not able to synthesize melanin, but capable of synthesizing tyrosinase, which results in lavender-brown skin color. Also referred to as T+.
Wild-type = The way the animal usually looks in nature (i.e. the normal color and pattern).
Xanthic = Having more yellow color than wild-type.

Description:

When you think of “Albino” you think of the classic Orange, Yellow and White albinos. Normal Albinos (T-) wipe out all of the Tyrosinase were as the Caramel Albinos (T+) allow some of the Tyrosinase enzyme to be processed creating the Caramel appearance. The only characteristic the Caramel Albino share with the “regular” Albino is the red irises.

Comments:

Caramel Albinos have been around since the mid 90’s, however, they are still rare in most collections. I purchased my first het Caramels in 1998 and in 2002 I produced a regular (T-) Albino which was totally unexpected. I have since outsourced different hets as well as a Caramel female in hopes of eventually producing Caramel Albinos. Hopefully this year (2005) I will produce a Caramel Albino from my original het female bred to a Pastel 100% het Caramel male. If not this will mean I haven’t been able to produce Caramels from het to het breeding out of 16 eggs. The odds of producing Caramels from het to het are 1 out of 4.

Update: As of March 2006 I have yet to produce a Caramel Albino.
